Asyntheticdual-functionoperonwithabistablestructurewasdesignedandsuccessfullyintegratedintothebacterialgenome. BistabilitywasgeneratedbythemutualinhibitorystructurecomprisedofthepromotersPtet andPlac andtherepressorsLacI andTetR.Dualfunctionessentialforcellgrowthwasintroducedbyreplacingthegenes(i.e.,hisCandleuB)encodingproteins involvedinthebiosynthesisofhistidineandleucinefromtheirnativechromosomallocationstothesyntheticoperon.Bothcolony formationandpopulationdynamicsofthecellscarrying thisoperonshowedthatthecellssurvivedstarvationandthenewly formedpopulationtransitedbetweenthetwostablestates,representingtheinducedhisCandleuBlevels,inaccordancewiththe nutritionalstatus.Theresultsstronglysuggestedthatthesyntheticdesignofproto-operonssensitivetoexternalperturbationsis practicalandfunctionalinnativecells. 1.Introduction nativegeneticstructuresandtoforcelivingorganismstouse foreignorprematuregeneticmodulestoachieveadaptat
