Metabolic ?ux analysis is a vital tool used to determine the ultimate output of cellular metabolism and thus detect biotechnologicallyrelevantbottlenecksinproductivity. (FBA)havemanypotentialapplicationsinbiotechnology.However,noteworthyhurdlesin?uxomicsstudyarestillpresent.First, severaltechnicaldi?cultiesinboth C-MFAandFBAseverelylimitthescopeof?uxomics?ndingsandtheapplicabilityof C-basedmetabolic?uxanalysis( C-MFA)and?uxbalanceanalysis 13 13 13 obtainedmetabolicinformation.Second,thecomplexityofmetabolicregulationposesagreatchallengeforprecisepredictionand analysisofmetabolicnetworks,astherearegapsbetween?uxomicsresultsandotheromicsstudies.Third,despiteidenti?ed metabolic bottlenecks or sources of host stress from product synthesis, it remains di?cult to overcome inherent metabolic robustness or to e?ciently import and express nonnative pathways. Fourth, product yields often decrease as the number of enzymatic steps increases.
