Genome-wideassociationstudieshavealsoidentifiedmultiplesinglenucleotidepolymorphisms(SNPs)intheIL23Rgene regionassociatedwithinflammatoryboweldiseases.ThisstudyexaminedtheassociationofIL23RandIL17AgeneSNPs withulcerativecolitissusceptibilityinapopulationinChina. Methodology:Atotalof270ulcerativecolitisand268healthycontrolswererecruitedfortheanalysesof23SNPsinthe IL23RandIL17Aregions.GenomicDNAwasextractedandanalysisofthese23SNPsusingligasedetectionreactionallelic (LDR)technology.GenotypeandalleleassociationswerecalculatedusingSPSS13.0softwarepackage. Principal Findings: Compared to the healthy controls, the variant alleles IL23R rs7530511, and rshowed a statistically significant difference for ulcerative colitis susceptibility (0.7% vs 3.3%, P=0.002; 60.4% vs 53.2%, P =0.0017, respectively).Thelinkagedisequilibrium(LD)patternsoftheseSNPsweremeasuredandthreeLDblocksfromtheSNPsof IL23RandoneblockfromthoseofIL17Awereidentified.Anovelassociationwithulcerativecolitissusceptibilityoccurredin haplotypesofIL23R(Block1H3P=0.02;Block2H2P=0.019;Block3H4P=0.029)andIL17A(H4P=0.034).Pair-wiseanalyses showedaninteractionbetweentheriskhaplotypesinIL23RandIL17A(P =0.014). Conclusions:Ourstudydemonstratedthatrs7530511,andrfIL23Rweresignificantlyassociatedwithulcerative colitissusceptibilityintheChinesepopulati
