Recently, in vitro studies revealedthattheactin-basedprocessivemotormyosinValsobindsto,anddiffusesalongmicrotubules.Thisbiophysical trickenablescellstoexploitbothtracksforthesametransportprocesswithoutswitchingmotors.Thedetailedmechanisms underlyingthisbehaviorremaintobesolved.BymeansofsinglemoleculeTotalInternalReflectionMicroscopy(TIRFM),we show here that electrostatic tethering between the positively charged loop 2 and the negatively charged C-terminal E- hooksofmicrotubulesisdispensable.Furthermore,ourdataindicatethatinadditiontocharge-chargeinteractions,other interactionforcessuchasnon-ionicattractionmightaccountformyosinVdiffusion.Thesefindingsprovideevidencefora novelwayofmyosintetheringtomicrotubulesthatdoesnotinterferewithotherE-hook-dependentprocesses.
