Methodology/PrincipalFindings:WeshowedherethatWnt5aactivatedDvl2,Daam1andRhoA,andpromotedmigration ofbreastcancercells,whichwas,however,abolishedbySecretedFrizzled-relatedprotein2(sFRP2)pretreatment.Dominant negative Dvl2 mutants or Dvl2 siRNA significantly decreased Wnt5a-induced Daam1/RhoA activation and cell migration. EctopicexpressionofN-Daam1,adominantnegativemutant,orDaam1siRNAremarkablyinhibitedWnt5a-inducedRhoA activation,stressfiberformationandcellmigration.EctopicexpressionofdominantnegativeRhoA(N19)orC3exoenzyme transferase,aRhoinhibitor,decreasedWnt5a-inducedstressfiberformationandcellmigration.
