Materials and Methods: The objective of the present study was to examine the impact of electrospun PLLA nanofiber scaffoldsonboneformationinvivo,usingacriticalsizeratcalvarialdefectmodel.Inadditionweanalyzedwhetherdirect incorporationofbonemorphogeneticprotein2(BMP-2)intonanofiberscouldenhancetheosteoinductivityofthescaffolds. Twocriticalsizecalvarialdefects(5mm)werecreatedintheparietalbonesofadultmaleSprague-Dawleyrats.Defectswere either(1)leftunfilled,ortreatedwith(2)bovinespongiosa,(3)PLLAscaffoldsaloneor(4)PLLA/BMP-2scaffolds.CranialCT- scans were taken at fixed intervals in vivo. Specimens obtained after euthanasia were processed for histology, histomorphometryandimmunostaining(Osteocalcin,BMP-2andSmad5). Results:PLLAscaffoldswerewellcolonizedwithcellsafterimplantation,butonlyshowedmarginalossification.PLLA/BMP-2 scaffoldsshowedmuchbetterboneregenerationandseveralossificationfociwereobservedthroughoutthedefect.PLLA/ BMP-2 scaffolds also stimulated significantly faster bone regeneration during the first eight weeks compared to bovine spongiosa.
