Methodology/PrincipalFindings:WegeneratedasolubleHA(sHA),derivedfromtheH3N2virusA/Aichi/2/68,modifiedat theC-terminuswithaGCN4pIItrimerizationrepeattostabilizethenativetrimericstructureofHA.Whenexpressedinthe baculovirussystem,themodifiedsHAformednativetrimers.Incontrast,theunmodifiedsHAwasfoundtopresentepitopes recognizedbyalow-pHconformationspecificmonoclonalantibody.Wefoundthatmiceprimedandboostedwith3m gof trimericsHAintheabsenceofadjuvantshadsignificantlyhigherIgGandHAItitersthanmicethatreceivedtheunmodified sHA. This correlated with an increased survival and reduced body weight loss following lethal challenge with mouse- adaptedA/Aichi/2/68virus.Inaddition,micereceivingasinglevaccinationofthetrimericsHAintheabsenceofadjuvants hadimprovedsurvivalandbodyweightlosscomparedtomicevaccinatedwiththeunmodifiedsHA. Conclusions/Significance:OurdataindicatethattherecombinanttrimericsHApresentsnativetrimericepitopeswhilethe unmodifiedsHApresentsepitopesnotexposedinthenativeHAmolecule.TheepitopespresentedintheunmodifiedsHA constitutea‘‘silentface’’whichmayskewtheantibodyresponsetoepitopesnotaccessibleinlivevirusatneutralpH.The resultsdemonstratethatthetrimericsHAisamoreeffectiveinfluenzavaccinecandidateandemphasizetheimportanceof structure-basedantigendesigninimprovingrecombinan
