The C. albicans ALS (agglutinin-like sequence) gene family encodes eight large cell-surface glycoproteins (Als1-Als7andAls9)thathaveadhesivefunction.ThisstudyutilizedC.albicansDalsmutantstrainstoinvestigatetheroleof theAlsfamilyinoralepithelialcelladhesionanddamage,cytokineinductionandactivationofaMAPK-based(MKP1/c-Fos) signalingpathwaythatdiscriminatesbetweenyeastandhyphae.OftheeightDalsmutantstested,onlytheDals3strain showedsignificantreductionsinoralepithelialcelladhesionanddamage,andcytokineproduction.Highfungal:epithelial cellmultiplicitiesofinfectionwereabletorescuethecelldamageandcytokineproductionphenotypes,demonstratingthe importanceoffungalburdeninmucosalinfections.Despiteitsadhesion,damageandcytokineinductionphenotypes,the Dals3straininducedMKP1phosphorylationandc-Fosproductiontoasimilarextentascontrolcells.Ourdatademonstrate thatAls3isinvolveddirectlyinepithelialadhesionbutindirectlyincelldamageandcytokineinduction,andisnotthefactor targetedbyoralepithelialcellstodiscriminatebetweentheyeastandhyphalformofC.albicans.
