A circulating recombinant form of HIV-1, CRF01_AE, is prevalent throughout Southeast Asia; however, only limited informationregardingtheimmunologicalcharacteristicsofCRF01_AEEnviscurrentlyavailable.Inthisstudy,weattempted toexaminetheevolutionarypatternofCRF01_AEEnvundertheselectionpressureofhostimmuneresponses. Methodology/PrincipalFindings:Peripheralbloodsampleswerecollectedperiodicallyover3yearsfrom15HIV-1-infected individuals residing in northern Thailand, and amplified env genes from the samples were subjected to computational analysis.TheV5regionofgp120showedhighestvariabilityinseveralsamplesover3years,whereastheV1/V2and/orV4 regionsofgp120alsoshowedhighvariabilityinmanysamples.Inaddition,theN-terminalpartoftheC3regionofgp120 showed highest amino acid diversity among the conserved regions of gp120. Chronological changes in the numbers of aminoacidresiduesingp120variableregionsandpotentialN-linkedglycosylation(PNLG)sitesareinvolvedinincreasing thevariabilityofEnvgp120.Furthermore,theC3regioncontainedseveralaminoacidres
