MolecularPhylogenyandDescriptionoftheNovel KatablepharidRoombiatruncatagen.etsp.nov.,and EstablishmentoftheHacrobiaTaxonnov NorikoOkamoto1,ChitchaiChantangsi2,Alesˇ Hora′k1,BrianS.Leander2,PatrickJ.Keeling1* 1Department of Botany, University of British Columbia, Vancouver, British Columbia, Canada, 2Departments of Botany and Zoology, University of British Columbia, Vancouver,BritishColumbia,Canada Abstract Background: Photosynthetic eukaryotes with a secondary plastid of red algal origin (cryptophytes, haptophytes, stramenopiles,dinoflagellates,andapicomplexans)arehypothesizedtoshareasingleoriginofplastidacquisitionaccording toChromalveolatehypothesis.Recentphylogenomicanalysessuggestthatphotosynthetic‘‘chromalveolates’’formalarge cladewithinclusionofseveralnon-photosyntheticprotistlineages.Katablepharidsareonesuchnon-photosyntheticlineage closely related to cryptophytes. Despite their evolutionary and ecological importance, katablepharids are poorly investigated. Methodology/PrincipalFindings:Here,wereportanewlydiscoveredflagellate,Roombiatruncatagen.etsp.nov.,thatis relatedtokatablepharids,butismorphologicallydistinctfromothermembersofthegroupinthefollowingways:(1)two flagellaemergefromapapilla-likesubapicalprotrusion,(2)conspicuousejectisomesarealignedinmultiple(5–11)rows,(3) eachejectisomeincreasesinsizetowardstheposteriorendoftherows,and(4)uponfeeding,apartofcytoplasmelastically stretch to engulf whole prey cell. Molecular phylogenies inferred from Hsp90, SSU rDNA, and LSU rDNA sequences consistentlyandstronglyshowR.truncataasthesisterlineagetoallotherkatablepharids,includinglineagesknownonly fromenvironmentalsequencesurveys.Acloseassociationbetweenkatablepharidsandcryptophyteswasalsorecoveredin most analyses. Katablepharids and cryptophytes are together part of a larger, more inclusive, group that also contains haptophytes, telonemids, centrohelids and perhaps biliphytes.
