Methodology/Principal Findings: In this study, 50 patients with esophageal squamous cell carcinoma (ESCC) were retrospectivelyanalyzed.OCT4wasexpressedin13cases(26%),andsurvivinwaspositivelyexpressedin31cases(62%), examinedbyimmunochemistry.OCT4wasfoundtobeanindependentpredictivefactorformediansurvivaltime,andthe patientsfromthesubgroupwithbothhighexpressionofOCT4andSurvivinhadtheworstprognosisinvestigatedbylog- ranktest.TofurtherexplorethemolecularregulatorymechanismbetweenOCT4andSurvivin,weconstructedthespecific smallhairpinRNA(shRNA)-expressingvectorstargetingOCT4or/andSurvivinandmanipulatedtheexpressionofOCT4and Survivin. By Western blotting and RT-PCR, we found that OCT4 could up-regulate Survivin expression in the esophageal cancercelllinesEca109andTE1.SimultaneouslyknockdownofOCT4andSurvivinexpressioninducedcellapoptosisand G2-phasedecreaseofcellcyclebyflowcytometry,andfinallyexertedanenhancedanti-proliferationpotencyinEca109and TE1celllinesbyMTTassay. Conclusions:ThisstudyshowsthatOCT4andSurvivinexpressionwerecorrelatedwithpoorsurvivalinpatientswithESCC. OCT4andSurvivinmayberegardedastargetsinESCCbiotherapy.
