Methods:Weinvestigatedplasmacytokinelevelsaswellasspontaneousandlipopolysaccharide(LPS)-stimulated cytokineproductionbyperipheralbloodmononuclearcells(PBMCs)inagroupof35severelytraumatizedPTSD patientscomparedto25healthycontrols. Results:Spontaneousproductionofinterleukin(IL)-1β,IL-6andtumornecrosisfactor(TNF)-αbyisolatedPBMCs wassignificantlyhigherinthePTSDcomparedtothecontrolgroupandevencorrelatedwithPTSDsymptom severitywithinthePTSDgroup.Incontrast,circulatingplasmalevelsofpro-andanti-inflammatorycytokinessuch asIL-6,IL-8,IL-10,TNF-α,ormonocytechemotacticprotein(MCP)-1werenotsignificantlyalteredinPTSDpatients comparedtohealthycontrols. Conclusions:OurfindingsindicatethatPBMCsofPTSDpatientsarealreadypre-activatedinvivo,providingfurther evidenceforlow-gradeinflammationinPTSD.Thismightpossiblyrepresentonepsychobiologicalpathwayfrom PTSDtopoorphysicalhealth.
