Rhamnose-InducibleGeneExpressioninListeria monocytogenes LarsFieseler.¤a,SibylleSchmitter.,JustinasTeiserskas¤b,MartinJ.Loessner* InstituteofFood,Nutrition,andHealth,ETHZurich,Zurich,Switzerland Abstract AcidproductionfromrhamnoseisacharacteristicphenotypeofListeriamonocytogenes.Wereporttheidentificationofthe rhamnosetransportandutilizationoperonlocatedatlmo2846tolmo2851,includingtherhamnose-dependentpromoter Prha.ExpressionofreportergenesundercontrolofPrhaonasinglecopyintegrationvectordemonstrateditssuitabilityfor induciblegeneexpressioninL.monocytogenes.TranscriptioninitiationfromPrhaisdosedependent,andaconcentrationas low as 100mM rhamnose was found sufficient for induction. Moreover, Prha is subject to glucose catabolite repression, whichprovidesadditionaloptionsforstrictcontrolofexpression.InfectionofhumanTHP1macrophagesrevealedthatPrha isrepressedinintracellularL.monocytogenes,whichisexplainedbytheabsenceofrhamnoseinthecytosolandpossible interferencebycataboliterepression.ThePrhapromoterprovidesanovelandusefultoolfortriggeringgeneexpressionin extracellularL.monocytogenes,whereasintracellularconditionspreventtranscriptionfromthispromoter.
