SpatialandTemporalLineageAnalysisofaPitx3-Driven Cre-RecombinaseKnock-InMouseModel MartenP.Smidt1,2*,LarsvonOerthel1,ElisaJ.Hoekstra1,2,RaymondD.Schellevis2, MarcoF.M.Hoekman1 1Molecular Neuroscience, Swammerdam Institute for Life Sciences, University of Amsterdam, Amsterdam, The Netherlands, 2Department of Neuroscience and Pharmacology,RudolfMagnusInstituteofNeuroscience,UniversityMedicalCenterUtrecht,Utrecht,TheNetherlands Abstract Developmentandfunctionofmesodiencephalicdopaminergic(mdDA)neuronshasreceivedalotofscientificinterestsince these neurons are critically involved in neurological diseases as Parkinson and psychiatric diseases as schizophrenia, depressionandattentiondeficithyperactivitydisorder(ADHD).Theunderstandingofthemolecularprocessesthatleadto normal development and function of mdDA neurons has provided insight in the pathology and provided critical informationonnewtreatmentparadigms.InordertobeabletostudyspecificgeneticablationinmdDAneuronsanew toolswasdevelopedthatdrivesCre-recombinaseunderthecontrolofthePitx3locus.ThePitx3geneiswellknownforits specificexpressioninmdDAneuronsandispresentattheonsetofterminaldifferentiation.Analysisofnewlygenerated Pitx3-Creknock-inmiceshowsthatCreexpression,measuredthroughtheactivationofeYfpbyremovalofa‘‘Stop’’signal (LoxP-Stop-LoxP-eYfpreportermouse),ispresentattheonsetofterminaldifferentiationandmimicscloselythenativePitx3 expressiondomain.Inconclusion,wepresenthereanewCre-drivermousemodeltobeusedintherestrictedablationof interestinggenesinmdDAneuronsinordertoimproveourunderstandingoftheunderlyingmolecularprogramming.
