Objective:TotestforassociationsbetweenvariantsinTSLP,TSLP-relatedgenes,andARinchildrenwithasthma. Methods:Wegenotyped15singlenucleotidepolymorphisms(SNPs)inTSLP,OX40L,IL7R,andRXRainthree independentcohorts:592asthmaticCostaRicanchildrenandtheirparents,422nuclearfamiliesofNorthAmerican childrenwithasthma,and239Swedishchildrenwithasthma.WetestedforassociationsbetweentheseSNPsand AR.Aswepreviouslyreportedsex-specificeffectsforTSLP,weperformedoverallandsex-stratifiedanalyses.We additionallyperformedsecondaryanalysesforgene-by-geneinteractions. Results:Acrossthethreecohorts,theTalleleofTSLPSNPrs1837253wasundertransmittedinboyswithARand asthmaascomparedtoboyswithasthmaalone.TheSNPwasassociatedwithreducedoddsforAR(oddsratios rangingfrom0.56to0.63,withcorrespondingFisher’scombinedPvalueof1.2×10-4).Ourfindingswere significantafteraccountingformultiplecomparisons.SNPsinOX40L,IL7R,andRXRawerenotconsistently associatedwithARinchildrenwithasthma.Therewerenominallysignificantinteractionsbetweengenepairs. Conclusions:TSLPSNPrs1837253isassociatedwithreducedoddsforARinboyswithasthma.Ourfindings supportaroleforTSLPinthepathogenesisofARinchildrenwithasthma.
