Pratietal.ArthritisResearchTherapy2012,14:R130 /content/14/3/R130 RESEARCH ARTICLE OpenAccess TreatmentwiththearginaseinhibitorNw- hydroxy-nor-L-argininerestoresendothelial functioninratadjuvant-inducedarthritis ClémentPrati1,2*,AlainBerthelot1,BernadetteKantelip3,DanielWendling2,4 andCélineDemougeot1 Abstract Introduction:Endothelialdysfunction(ED)participatestoatherogenesisassociatedtorheumatoidarthritis.We recentlyreportedincreasedarginaseactivity/expressioninvesselsfromadjuvant-inducedarthritis(AIA)rats.Inthe presentstudy,weinvestigatedtheeffectsofacurativetreatmentwiththearginaseinhibitorNw-hydroxy-nor-L- arginine(nor-NOHA)onvasculardysfunctioninAIArats. Methods:AIAratsweretreatedwithnor-NOHA(40mg/kg/d,ip)for21daysaftertheonsetofarthritis.Agroupof untreatedAIAratsandagroupofhealthyratsservedascontrols.EDwasassessedbythevasodilatoryeffectof acetylcholine(Ach)onaorticrings.Theroleofsuperoxideanions,prostanoids,endothelium-derivedhyperpolarizing factor(EDHF)andnitricoxidesynthase(NOS)pathwaywasstudied.PlasmalevelsofIL-6andvascularendothelial growthfactor(VEGF)weredeterminedbyELISAkits.Arthritisseveritywasestimatedbyaclinical,radiologicaland histologicalanalysis. Results:Nor-NOHAtreatmentfullyrestoredtheaorticresponsetoAchtothatofhealthycontrols.Theresults showedthatthisbeneficialeffectismediatedbyanincreaseinNOSactivityandEDHFandreducedsuperoxide anionproductionaswellasadecreaseintheactivityofcyclooxygenase(COX)-2,thromboxaneandprostacyclins synthases.Inaddition,nor-NOHAdecreasedIL-6andVEGFplasmalevelsinAIArats.Bycontrast,thetreatmentdid notmodifyarthritisseverityinAIArats. Conclusions:ThetreatmentwithanarginaseinhibitorhasapotenteffectonEDinAIAindependentlyofthe severityofthedisease.Ourresultssuggestthatthisnewpharmacologicalapproachhasthepotentialasanovel add-ontherapyinthetreatmentofRA.
