Methods:WeevaluatedtheexpressionofTCTPatbothRNAmessangerandproteinlevelbyusingreverse transcriptaseanalysis,immunoblottingandimmunohistochemistryin10healthysamplescornea:fourobtainedafter penetratingkeratoplastyandsixfromeyesenucleatedforotherpathologies.Finally,weanalysedby immunohistochemistrytenparaffin-embeddedsamplesofHerpessimplexviruskeratitiscollectedatSiena DepartmentofHumanPathologyandOncology:5hadclinicallyquiescentdiseaseand5hadactivecorneal inflammation. Results:ReversetranscriptaseandimmunoblottingdemonstratedTCTPexpressionincorneaasa22,000Da molecularweightbandcorrespondingtothemolecularweightofthisprotein.Immunohistochemically,allthe layersofnormalcornealepitheliumshowedTCTPcytoplasmicexpression.TCTPwas,also,observedinkeratocytes andintheendothelium.InHerpessimplexviruskeratitissamples,strongexpressionofTCTPwasevidentinstromal cells,intheinflammatoryinfiltrateandinneo-vessels. Conclusions:Inthispreliminarystudywedemonstrated,forthefirsttime,thepresenceofTCTPinhumancornea, suggestingapotentialroleinthepathogenesisofherpesviruskeratitis.
