Methods:Blgdigestawerepreparedusinganinvitrodigestionprotocolsimulatingeithergastricdigestionalone orfollowedbyduodenaldigestionwithorwithoutphosphatidylcholine(PC).Biochemicalanalysisofblgdigesta wasperformedbySDSandtheirconcentrationwasmeasuredbyasandwichELISA.Assessmentoftheir allergenicreactivitywasdoneinvitrobyEASTinhibition,specificbasophilactivation(basotest)andlymphocyte proliferation(PCNA-flowcytometry)assaysusingseraandcellsfrompatientsallergictoblgandinvivobyskin pricktesting(SPT)ofthesepatients. Results:Blgwasonlybrokendowntosmallerpeptidesaftergastro-duodenaldigestionalthoughasizeable amountofintactproteinstillremained.DigestiondidnotmodifytheIgEbindingcapacityofblgexceptforgastro- duodenaldigestionperformedintheabsenceofPC.Theseresultsareconsistentwiththequantityofintactblg remaininginthedigesta.Overallbothgastricandgastroduodenaldigestionenhancedactivationofsensitized basophilsandproliferationofsensitizedlymphocytesbyblg.However,therewasatendencytowardsreductionin meandiameterofSPTfollowingdigestion,thePCaloneduringphase1digestioncausingasignificantincreasein meandiameter.
