Abstract
Background:ThefelineendogenousretrovirusRD114iscontainedinthegenomeofcats.Thevirusmay
contaminatelivecaninevaccinesbasedonculturedfelinecells.Theinvivoinfectivity,acuteandsubacute
pathogenicity,andviralproliferationoftheRD114viruswereevaluatedbyexperimentalinfectionofdogs.
Methods:Ninespecificpathogenfreedogsweredividedintothreegroups,witheachgroupconsistingofone
femaleandtwomaledogs.Dogsweresubcutaneouslyinoculatedintheneckwitheither1mlRD114stockvirus
(groupA),inactivatedRD114virussuspension(groupB),orcellculturemedium(groupC)asanegativecontrol.To
assessbloodcellcountsandbiochemicalproperties,bloodsamplesfromeachgroupwerecollected5daysbefore
inoculation,justpriortoinoculation,and1,3,7and10dayspost-inoculation.
Result:Duringtheexperimentalperiodof51days,noneofthedogsinoculatedwithRD114virusshowedany
clinicalsigns,significantincreasesinrectaltemperatureorabnormalbloodbiochemicalcharacteristicsincluding
C-reactiveproteinwhencomparedwiththenegativecontrols.Wewerenotabletore-isolatetheRD114virusfrom
buffycoatcellsofgroupAdogs.Additionally,wecouldnotdetectRD114provirusinthegenomicDNAisolated
fromperipheralbloodleukocytes,lymphnode,spleenandsternalbonemarrowcells.
Conclusions:SignsofRD114virusproliferationwerenotfoundaftersubcutaneousinfectionofdogs.Althoughthe
potentialriskcausedbyinfectionwithRD114virusindogscouldnotbeassessedinthisstudy,wesuspectthat
RD114virushaslittleornovirulenceindogs.
