中药夏枯草对甲状腺癌细胞NIS基因表达及摄碘率影响 　 作者：张王峰, 付强, 赵华栋, 徐海峰，周润锁, 袁梦晖, 周飞华, 南菁 【摘要】 目的： 探讨中药夏枯草对甲状腺癌细胞株K1钠/碘同向转运体(NIS)基因表达及摄碘率的 影响 ，为夏枯草辅助放射性碘 治疗 甲状腺癌提供依据. 方法 ： 分别以不同浓度(0, 20, 50, 100, 150, 200 g/L)的夏枯草处理体外培养的甲状腺癌细胞株(K1)，48 h后利用半定量RTPCR检测细胞NIS mRNA表达，γ计数仪检测细胞摄碘能力. 结果： 夏枯草浓度在0～100 g/L范围内，细胞NIS基因表达及摄碘能力随夏枯草浓度的增加而增加(Plt;0.05). 当夏枯草浓度达150～200 g/L时，增加后浓度与100 g/L浓度相比差别无统计学意义(Pgt;0.05). 结论： 夏枯草可上调甲状腺癌K1细胞NIS基因表达，增强其摄碘率，而且这种作用在一定浓度范围内具有剂量依赖性. 【关键词】 夏枯草；甲状腺肿瘤；肿瘤细胞；RTPCR；碘同向转运体；摄碘率 　　【Abstract】 AIM: To investigate the effect of traditional Chinese medicine Prunella vulgaris on the expression of sodium iodide symporter (NIS) gene and radioiodide uptake in thyroid cancer cell line(K1), so as to evaluate the significance of traditional Chinese medicine Prunella vulgaris in the treatment of thyroid cancer with radioiodide. METHODS： The thyroid cancer cell line(K1) was treated with traditional Chinese medicine Prunella vulgaris at concentrations of 0, 20, 50, 100, 150, 200 g/L for 48 h. The expression level of NIS mRNA of K1 was determined by RTPCR and its radioiodide concentration activity was measured by γcounter. RESULTS： Traditional Chinese medicine Prunella vulgaris at the concentration between 0 and 100 g/L increased the expression of NIS gene and the radioiodide uptake of K1.
