The existence, th ickness and chem i cal constituents of the capsu le are mi portant characters of acteria. Capsu le stain ing and o serving are asic skills in acterial certification. It needs proper staining methods and proficiency to m ake stain ing slide of capsule etter, ecause the capsule has low refraction and low affinity w ith dyes. B ac illus m uc i lag inosus is characterized y its a undant capsu le, and m ost useful perform ances of the acteria relate to its capsu les. W e stud ied seven m ethods of negative stain ing of acterial capsule and compared their ad vantages and w eaknesses.
