添加氧载体对黄原胶发酵的影响 程蓉,张永奎 (四川大学化学工程学院,四川成都 610065 ) 摘要:对加入氧载体的黄原胶发酵体系进行了研究。实验结果表明,大豆油与正己烷是两种 有效的氧载体;在发酵初期添加6 % (v/v )的大豆油,黄原胶产率可达3.02 %,在发酵12 h 添加1 % (v/v )的正己烷,产率可达2.95 % ,与对照组相比产率分别提高89 %与84 %;添 加正己烷后,整个发酵过程的pH 值均偏高,推测在发酵体系中,正己烷不仅起到氧载体的 作用,也对细菌细胞膜的通透性产生影响,改变了细胞与环境进行物质交换的过程。氧载体 的添加对黄原胶的质量影响不大,在加入大豆油和正己烷后,黄原胶丙酮酸含量分别为3.92 %和3.18 % (对照组为3.33 % )。 关键词:黄原胶;氧载体;好氧发酵;正己烷;大豆油 中图分类号:TS202.3 ;TS201.2 Influences of oxygen vectors on the production of xanthan gum CHENG Rong, LIU Chen-lin, WEN Hao-cheng, YANG Jun, YANG Qiu-yan (Chemical Engineering Institute, Sichuan University, Chengdu 610065, China) Abstract: Xanthan gum fermentation is strongly dependent on the dissolved oxygen concentration. The system with the addition of oxygen vector was applied to enhance oxygen transfer. The addition of oxygen carriers (soybean oil, n-hexane) obviously improved xanthan gum yield. When added 6 % (v/v) soybean oil at the initial stage of fermentation and 1 % (v/v) n-hexane after 12 h of culture, the results were 3.02 % and 2.95 % for the gum productivity respectively (improved by 89 % and 84 %, respectively). Through fermentation with addition of 1 % (v/v) n-hexane, the broth showed a relative high pH tendency to the other two groups. The results suggested that Xanthomonas campestris ’ cell membrane were influenced, which had an important impress on the transport of former substantial for the biosynthesis of xanthan gum. The quility of xanthan gum was not influenced significantly, and the pyruvate content of xanthan with addition of soybean oil and n-hexane were 3.92 % and 3.18 % respectively (the control: 3.33 %).
