AbstractInvitroevaluationofdendriticcellstargetingthermo-sensitivedeliverysystemforsiRNAInvitroevaluationof dendriticcellstargeting thermo-sensitivedelivery systemforsiRNA AbstractDestination:InhibitingtheExpressionofapoptosisgeneBakindendriticcellsbyusingsiRNAinterferencetechnologyandevaluatingoftheinhibitoryeffectatthecellular level.But,thereexistsomeshortagesofsiRNAindrugdeliverysystem.Forexamplelow targeting,lowtransfectionefficiencyandinstabilityinvivoandsoon.So,weshould designanefficientsiRNAdeliverysystemtomakesurethesiRNAtargetingBakgenein DCs.ThereforetheresearchofsiRNAdeliverysystemtargetingBakwillbeexpectedto increaseitsantigenpresenting,promotetheimmuneresponseandenhanceanti-tumor effects.Based onthesurfaceexpressionofDEC-205receptorondendriticcells,the DEC-205modifiedpoloxamer-g-chitosancontainingsiRNAtargetingBakwassynthesized topreparethermo-sensitivemicelles,andthenano-carrierswerethencharacterizedwith respecttothepharmaceuticalproperties,cytotoxicity,targeting,uptake rateandmechanism, endosomeescapeandsilencingefficiencyofsiRNAandsurvivalrateofDCs.Giving theoryaccordingstobuildsafeandeffectivedurgdeliverysystemofantitumorsiRNA vaccine.Method:First,synthesisofDEC-205modifiedpoloxamer-g-chitosanbasedonthe previousworkinourlaboratoryandsiRNAloadedPMswerepreparedbydirectly dissolvedmethod.ThecharacteristicsofPMsonsize,Zetapotential,encapsulation efficiency,thermo-sensitive,stabilityandcytotoxicitywereinvestigated.DCswere generatedfrommurinebonemarrowcellbyanestablishedinvitromethod.Lipofectamine 2000asthepositivecontrol,subsequently,theuptakenofsiRNAloadedDEC-205-Po-CS byDCswasmeasuredusingflowcytometry.Theendocytosisandendocytosispathof micelleswasexpalnedbyatomicforcemicroscopeandendocytosisinhibitors.The morphologyofthecellendosomeswithcoldshockwasobservedbyfluorescence microscope.Eventually,theexpressionofBakinDCswasdetectedwithWesternblotand RT-PCR,whilethesurvivalrateofDCsaftertransfectionofBaksiRNAloaded nanomicells wasmeasuredbycelldeathdetectio
