SilicSilicon-based Raman platform for cellular analysis and detectionAbstractSilicon-based Raman platform for cellular analysis anddetectionAbstractSilicon nanomaterials are a type of important semiconductor nanomaterials with attractive properties including excellent electronic/mechanical properties, favorable biocompatibility, huge surface-to-volume ratios, surface tailorability, as well as their compatibility with conventional silicon technology. Consequently, there has been great interest in developing functional silicon nanomaterials for various applications ranging from electronics to biology. In this paper, we develope a silicon-based surface-enhanced Raman Scattering (SERS) strategy for apoptosis detection at the single-cell level;afterwards,we present a silicon-based Raman mapping strategy for DNA-related in vitro studies. The details are as follows,1. We report a kind of silicon-based SERS-active and reproducible substrate, i.e., silver nanoparticles decorated-silicon wafers (AgNPs@Si), SERS in vitro sensing strategy for sensitive detection of apoptotic cells. Remarkably, single-cell detection of different apoptotic cells is readily realized using the AgNPs@Si of high SERS sensitivity and excellent reproducibility. In addition, the detection process is label-free; i.e., DNA intrinsic SERS spectra are directly employed for apoptosis detection, which is free of additional chemical or biological reagents as external signal indicator. Consequently, our SERS method may serve as a potentially practical tool for ultrasensitive detection of apoptotic cells, complementing the state of-the-art strategies for apoptosis detection.The present findings may open new opportunities for SERS-based in vitro sensing applications.2.we develop a Si-based Raman mapping strategy, which is based on six-color cellular Raman mapping images constructed from the intensities of Si intrinsic Raman scattering peak at ~520 cm-1.
