SectionⅡTheeffectoftotalglycosidesofranunculusjaponiucusoncalciumionslevelinvascularsmoothmusclecellsofratsObjective:Toobservedtheeffectoftotalglycosidesofranunculusjaponiucusoncalciumionslevelinvascularsmoothmusclecells.Method:WeusedanewgenerationsofCa2+fluorescentprobe(Fluo-3/AM)tomarkcalciuminvascularsmoothmusclecellswhichwecultured.Observedthefluorescenceimagingbyuseinvertedfluorescencemicroscopeandmeasuredthefluorescenceintensityofcalciuminvascularsmoothmusclecellsbyusefluorescencespectrophotometer.Toobservedtheeffectoftotalglycosidesofranunculusjaponiucusoncalciumionslevelinvascularsmoothmusclecellsofrats.Results:TheangiotensinⅡcansignificantlypromotethecalciumionsinextracellularinnerflow.ThedifferentdosesoftotalglycosidesofranunculusjaponiucuscanreducethecalciumionslevelincellswhichmediatedbyangiotensinⅡ.Theresultsisrelatedthedosesoftotalglycosidesofranunculusjaponiucus.Conclusion:TotalglycosidesofranunculusjaponiucuscanantagonizetheeffectofAngⅡwhichcansignificantlypromotethecalciumionsinextracellularinnerflowpartⅡTheeffectivepartoftotalglycosidesofranunculusjaponiucusObjective:Tostudytheeffectpartoftotalglycosidesofranunculusjaponiucusonvasodilatationinvitro.Method:Werecordedthetensionofthoracicaorticinvitroandobservedtheinhibitioneffectofdifferentpartsoftotalglycosidesofranunculusjaponiucusoncontractionofthoracicaorticwhichmediatedbynorepinephrine(NE)andKCI.AstheRutintothestandardanduseUVspectrophotometertomeasurethetotalflavonoidcontentindifferentpartsoftotalglycosidesofranunculusjaponiucus.MakethecorrelationregressionoftheflavonoidcontentasabscissaandtheinhibitionratesasverticalcoordinatebyLogisticandlinearstatistics.Results:(1)ThedifferentalcoholpartoftotalglycosidesofranunculusjaponiucuscaninhibitionthetensionincreasedofthoracicaorticinvaryingdegreeswhichmediatedbynorepinephrineandKCI(P0.05).(2)Full-wavelengthscanningtodeterminethemaximumabsorptionwavelengthis358nm.TherelativestandarddeviationprecisionofexperimentsandstabilityexperimentRSDis1.00%.Theave
