investigatedbyCell-Matrigeladhesionexperiment.Matrigelcanbere-formatedintheculturemediumwhichisverysimilartothenaturalbasementmembrane.Results:1.SettingupthespontaneousandexperimentallungmetastasismodelofB16melanoma.After8to12days,themelaninspotcameoutintheinoculationsite.Thenoduleswascomeintobeingwhenthemelaninspotgraduallyincreased.HEstaininghistologicalexamination:Thestructureoftumortissuewasnestedordispersed.Largecellvolume,polygon,nuclearvacuolization,prominentnucleoli,somevisiblemitotic.2.After23daysthemodelsweresetup,thetumor-bearingclawswereresectedandweighedup.PNScouldinhibitthegrowthofB16melanoma.Theinhibitionrateofthe480mg·kg-1PNSgroupwas50.85%afterstatisticalanalysisoftumorweightdata.3.After46daysthemodelsweresetup,themicewerekilled,removedthelungstoobservethelungtumormetastasis.PNScanobviouslyinhibitthelungmetastasisofB16melanoma.Thenumberandsizeoftumorcoloniesinlungofthe240mg·kg-1PNSgroupand480mg·kg-1PNSgroupwerelowerthanthatinthenegativecontrolgroup.4.Theexpressionofconnexin32inmelanomaweredetectedbyimmunohistochemistry:PNScouldup-regulatedtheexpressionofconnexin32inmembraneofmelanoma.5.TheGJICofB16cellsindifferenttreatmentgroupsweredetectedbySL/DT:Inthenegativecontrolgroup,theLuciferyellowwaslimitedtotheprimaryscrapedcellswithnosignificantLuciferyellowtranfer,andGJICwasnegative.AftertreatedwithPNS（100μg·ml-1、200μg·ml-1、400μg·ml-1）,thetransferringrangeofLuciferyellowincreasedgradually.TheGJICfunctionofB16cellshavebeeneffectivelyrestored.6.TheexpressionofCx32indifferenttreatmentgroupsweredetectedbywesternblot:PNScouldnotincreasethequantityoftheCx32proteininB16cells.7.Cell-Matrigeladhesionexperiment:TherelativeratesoftumorcelladhesionindicatethatPNShasinhibitoryeffectontheadhesioncapacityofB16cellswithMatrigel.Andalltrans-retinoicacid（RA）hasthesameeffectasareinforcingagent.Conclusion:PNScouldinhibitmalignantmelanomagrowthandmetastasis.PNScouldup-regulatedtheexpressionofconnexin32inmembraneofmelanomaandtheGJICfunctionofB16cellsbutitcouldnotincreasethequa
