CCL18结合CCR8受体对膀胱癌5637细胞增殖和侵袭影响 　　【摘要】 目的：探?CCL18与CCR8受体结合对膀胱癌5637细胞增殖和侵袭的影响，为抑制膀胱癌迁移提供实验基础。方法：检测不同浓度CCL18下膀胱癌细胞中EMT部分标志物的蛋白表达变化。通过转染siRNA沉默CCL18的受体CCR8以研究对膀胱癌的生物学行为的影响及产生的相关变化，并进一步观察细胞形态、侵袭能力、肿瘤转移的相关变化情况。结果：在不同浓度CCL18处理下，E-cadherin随CCL18浓度增加其蛋白表达下调，而MMP-2、VEGF随CCL18浓度增加其蛋白表达上调。沉默CCR8受体后检测膀胱癌细胞连接变松散，细胞形态相对较圆；CCR8、MMP-2、VEGF-C的mRNA及蛋白表达水平下调，E-cadherin的mRNA及蛋白表达水平上调；transwell结果（200倍镜下）显示膀胱癌细胞数显著减少，差异均有统计学意义（P0.05）。结论：CCL18通过结合CCR8受体促进膀胱癌细胞的增殖与侵袭。 　　【关键词】 膀胱癌； CCL18； CCR8受体； 细胞增殖 　　Effect of CCL18 Combined with CCR8 Receptor on Proliferation and Invasion of Bladder Cancer Cell Line 5637/CHENG Xinfu，FU Bin.//Medical Innovation of China，2017，14（35）：028-032 　　【Abstract】 Objective：To investigate the effect of CCL18 combined with CCR8 receptor on proliferation and invasion of bladder cancer cell line 5637，and to provide an experimental basis for the inhibition of bladder cancer migration.Method：The expression of some EMT markers in bladder cancer cells under different concentrations of CCL18 was detected.Changes by transfection of siRNA silencing CCL18 receptor CCR8 in order to study the biological behavior of bladder cancer and the further observation of cell morphology and invasion related changes，tumor metastasis.Result：At different concentrations of CCL18，the expression of E-cadherin was down regulated with the increase of CCL18 concentration，while the expression of MMP-2 and VEGF was up-regulated with the increase of CCL18 concentration.Loose connection detection of bladder cancer cells after silencing CCR8 receptor，cell morphology was relatively round.mRNA and protein expression of CCR8，MMP-2 and VEGF-C levels were down regulated expression of mRNA and E-cadherin protein level increased.Transwell results（200 magnification） showed that bladder cancer cell number was significantly reduced，the differences were statistically significant（P0.05）.Conclusion：CCL18 combined with CCR8 receptor can promote the proliferation and invasion of bladder cancer cells. 　　【Key words
