Abstract Shrimp aquaculture has become a mainstay aquaculture industry in China. But in recent years, the shrimp industry has suffered from huge economic loss with the infectious diseases and biosafety. Therefore, researchers suggested that the use of the ecology control method of Give priority to immunological control, combining prevention and treatment and the exploration of the green antibacterial peptide instead of antibiotics will be very helpful to solve these problems. Interestingly, recently it was demonstrated that some small molecular fragments derived from hemocyanin existing in the shrimp haemolymph possessed antibacterial activities. However, so far the research of the fragments is not comprehensive. Thus, in the present study, an attempt was made to indentify the hemocyanin-derived fragments from shrimp Litopenaeus vannamei and investigate their antibacterial activities and related-degradation enzymes. The main results are as follows: Prediction and antimicrobial activities analysis of the hemocyanin-derived fragments Three online prediction softwares (namely, AntiBP Server, CAMP, and APD2) was carried out to predict the hemocyanin-derived fragments of L. vannamei. The results indicated that hemocyanin might produce 34 antibacterial peptide fragments with the molecular weight ranged from 1.5 to 1.9 kDa. Of these, 12 fragments might contain an alpha helix structure and interact with bacteria, suggesting that they are very likely to act as antibacterial peptides. Furthermore, the 12 peptide fragments were synthesized by using the method of solid-phase peptide synthesis, and then analyzed their antibacterial activities. The results showed that these synthetic peptides revealed obvious antibacterial activities against beta-Streptococcus and (or) V. parahaemolyticus, and the antibacterial activities of them were more than 90% compared to that of control group.
