MyoD1、myogenin在儿童横纹肌肉瘤中的表达 及其临床意义?基金项目： ?基金项目：重庆医科大学校级课题重点项目：儿童常见实体恶性肿瘤诊断和治疗相关基因筛选的研究（XBZD200708） 作者单位：重庆医科大学附属儿童医院肿瘤外科 作者简介：马玉虹 女 硕士 电话E-mail： HYPERLINK mailto:kukumalu122@126.com kukumalu122@126.com 通信作者：王珊 教授 E-mail：wangshan778@163.com 马玉虹，王珊 ，邓晓斌，李想 (重庆医科大学附属儿童医院肿瘤外科，重庆400014) 摘 要：目的 探讨MyoD1、myogenin在儿童横纹肌肉瘤及其他三种儿童小细胞恶性肿瘤中的表达情况及其临床意义。方法 选取胚胎型横纹肌肉瘤11例，腺泡型横纹肌肉瘤5例，神经母细胞瘤、恶性淋巴瘤、尤文肉瘤各10例，行HE染色，并应用S-P免疫组织化学染色法检测MyoD1、myogenin蛋白在这四种儿童小细胞恶性肿瘤中的表达水平。结果 MyoD1、myogenin在横纹肌肉瘤中阳性表达率分别为：75%，68.75%，在其他三种肿瘤中阳性表达率均为0%，MyoD1、myogenin在横纹肌肉瘤组中的表达与其他肿瘤组比较有显著差异（P0.05），两种蛋白在腺泡型横纹肌肉瘤中的阳性表达率明显高于胚胎型横纹肌肉瘤（P0.05）。肿瘤恶性程度越高，MyoD1和myogenin阳性表达就越强（P0.05）。MyoD1、myogenin表达与儿童横纹肌肉瘤预后有明显的相关性，两种蛋白在预后差组中的阳性表达率明显高于预后中组（P0.05）。结论 MyoD1、myogenin可以作为辅助鉴别横纹肌肉瘤及其他小细胞恶性肿瘤的依据；MyoD1、myogenin基因可以作为辅助诊断儿童横纹肌肉瘤的综合指标，也可作为鉴别儿童胚胎型横纹肌肉瘤和腺泡型横纹肌肉瘤的有用的分子生物学标志；检测儿童横纹肌肉瘤中MyoD1、myogenin的表达情况对横纹肌肉瘤恶性程度及预后的判断有一定的临床指导意义。 关键词：横纹肌肉瘤；儿童；MyoD1；myogenin The expressions and clinical significances of MyoD1 and myogenin in rhabdomyosarcomas of childhood Ma Yuhong, Wang Shan，Deng Xiaobin，Li Xiang（Surgical Oncology，Childrens Hospital of Chongqing Medical University, Chongqing 400014，China） ABSTRACT：Objective To investigate the expression of MyoD1、Myogenin in rhabdomyosarcoma（RMS）and other small cell malignant tumors of childhood，and approach the clinical significance. Methods To collect 11 cases of Embryonal rhabdomyosarcoma（ERMS）, 5 cases of alveolar rhabdomyosarcoma（ARMS），10 cases of neuroblastoma，10 case of malignant lymphoma and 10 case of Ewings sarcoma），dyeing by HE ，and to detect the expression levels of the proteins of MyoD1 and Myogenin by S-P immunohistochemistry method in these 4 types of small cell malignant tumors.. Results The positive expressions of MyoD1 and myogenin in RMS are 75% and 68.75%，but the expression rates of other 3 types of small cell malignant tumors are 0%. The positive expression rate and intensity of MyoD1 and myogenin in RMS group are higher than them in other 3 types of small cell
