延边大学临床医学院内科学硕士论文THE 延边大学临床医学院内科学硕士论文 THE EFFECTS OF CHRONIC ALCHOLISM AND WITHDRAWAL ON LEARNING AND MEMEoRY AND NMDA RECEPToR SUBUNITS NMDAR2B EXPRESSl0N IN THE HIPPOCAMPUS OF RATS Abstract objective：To study the effects of chronic alcoholism and withdmwal on learning and memory and NMDA receptor subunits NMDAR2B expression in hippocampus ofrats， Methods：Fifty—eight wistar male rats were randomly divided into control group and experimental group，and the experimental group were divided into given 1 5％alcohol group，given 50％alchol not withdrawal group，withdrawal 36h group，withdrawal 3d group，withdrawal lw group and withdrawal 2w group．Experimental group were poured into 50％or 15％alcohol 10ml／kg／d，and control group were poured into physiological saline of sanle quantity for 8 weeks．After completion of pouring the alcohol to experimental rats，all groups of rats were tested by water maze．Afterwards，the brain tissue sections were performed by immunohistochemistry to demonstrate the expression of NMDAR2B imnmnoreactivity in the hippocampal CAI，CA3 and dentate gyms then they were analyzed by computer-assisted image analysis system． Results：(1)Withdrawal earl be observed in the given 50％alcohol group at the eighth week；(2)by comparing with the rats of control group and the given 1 5％alcohol group． the latent periods to search platform was prolonged in the given 50％alcohol group and the 2w withdrawal group；(3)by comparing with the control group the expression of NMDAR2B WaS significantly up—regulated in the given 50％alcohol group and the given 1 5％alcohol group；(4)by comparing with the rats of control group，the expression of NMDAR2B was significantly up-regulated in the 36h，3d and lw withdrawal group and got to maxium at the 3th day then decresed graduately from lw；(5)by comparing with the mrs of control group，the expression of NMDA2B was significantly down-regulated in the 2w withdrawal group． Conclusion：Chronic alcoholism might decrease the ability of learning and memory；
