植物资源与环境学报 2006，15(2)：50—54 Journal of Plant Resources and Environment 太子参快繁技术的优化及同源四倍体的诱导与鉴定 谢 燕，高山林 ，曾 杨，王 蔚，刘 蓁 (中国药科大学，江苏南京210038) 摘要：以太子参[Pseudostellaria heterophylla(Miq．)Pax]二倍体组培苗为实验材料，运用正交实验设计和组织培养 方法，对太子参试管苗快速繁殖技术进行优化，并进行了同源四倍体的诱导与鉴定。结果表明，太子参最佳的繁殖 培养基为含1．0 mg·L 6一BA和0．2 mg·L NAA的MS培养基；最佳生根培养基为含0．1 mg·L～IAA、0．2 mg·L NAA和1．0 mg·L ABT或0．2 mg·L IAA、0．2 mg·L NAA和1．0 mg·L ABT的1／2MS培养基。 诱导同源四倍体的最佳处理方法为：用0．2％秋水仙素处理22 h或用0．3％秋水仙素处理12 h，所诱导的同源泉四 倍体染色体数目为2n：4x=64条。 关键词：太子参；快繁；多倍体；诱导；鉴定 中图分类号：$567．5 3．035 文献标识码：A 文章编号：1004—0978(2006)02—0050—05 The optimization of rapid propagation technique and the induction and identification of autotetraploid ofPseudostellaria heterophylla XIE Yan，GAO Shan—lina~ ZENG Yang，WANG Wei， ， LIU Zhen(China Pharmaceutical University，Nanjing 210038，China)，J．Plant Resour．Environ． 20o6．15(2)：50—54 Abstract：The method of rapid propagation in tissue culture of Pseudostellaria heterophylla(Miq．)Pax was optimized by using orthogonal experiment．The autotetraploid induction and identification were carried out n vitro． The results indieated that the best multiplying medium was MS containing 1．0 mg·L 6一BA and 0．2 mg·L NAA．The strong root was induced on the 1／2 MS medium containing 0．2 mg·L NAA．1．0 mg·L ABT and 0．1 or 0．2 mg·L IAA．The autotetraploid strains could be induced by immersing buds in 0．2％ colchicine solution for 22 h or in 0．3％ colchicine solution for 12 h．The chromosome number of autotetraploid strain was 2n=4x=64． Key words：Pseudostellaria heterophylla(Miq．)Pax；rapid propagation；autotetraploid；inducing； identificati0n 太子参是石竹科植物孩儿参(PseudosteUaria heterophylla(Miq．)Pax]的干燥块根，有益气健脾、 1 材料与方法 生津润肺等功效；主治脾胃虚
