华中农业大学硕士论叉Abstract 华中农业大学硕士论叉 Abstract The work Dresents the validation ofan RP·HPLC method with UV detection at 287nm fbr the auantitation determination of lomefloxacin in plasma，skin，muscle and hepato．．pancreas of common carps(Cyprinus carpio)．Chromatography seperation was achieved on a ODS Hypersil—C18(1 50 X 4．6mm，5 p m)column using a mixature of0．05M potassium dihydrpgen phosphate，methanol and 0．5M tetrabulyl ammonium bromide(v／v／v 85：1 5：4)as the mobile phase．Lomefloxacin was extracted from plasma and tissues with dichloromethane at pH7．4，and the extracts were evapormed to dryness at 50。C water bath，dissolved in mobile phase after clean·up procedure by hexane and injected into the HPLC system with 20 u 1．Calibration graphs were prepared for a11 samples and good tinearity was achieved over the concentration(0．1-20 u g／ml(g))ranges tested(r0．9996)and goodness offit12％。Mean recoveries were about75％． The pharmacokinetic disposition of lomefloxacin was described after a single oral administration of 20rag．kg1．bw-1 dose in seventy healthy common carps(Cyprtnus carpio)．Lomofloxacin concentration in the plasma was determined by high-performance liquid chromatography(HPLC)with a limit of detection of 5ug／L in plasma．The pharmacokinetic profiles following oral administration were determined utilizing a classical linear tow—compartment open model．The results indicated that the apparent volume of distribution(Vd／F)，elimination half-life(t xn 9)and absorption halfilife (tl，2t、a)，were 5．21L／kg，20．03h and O，31h，respectively,the peak of plasma concentration (CⅢax)，the time to reach Cm“(tp)and the area under the curve(AUC)were 1 1．20 u g／mL．0．45h and 1 1 O．93 u g／mL’h．A HPLC method was developed for determination of tissues residues of lomefloxacin in common carps(Cyprinus carpio)．After a single oral administration ofa dosage of20mg．kg-I．bwl，lomefloxacin was detected in ld，3d，5d，7d， 9d，1 2d．Lomefloxacin could be detected in muscle，plasma，skin and hepato-pancreas I 2 days after
