※分析检测 食品科学 2009, Vol. 30, No. 12 233 免疫捕捉- 通用引物PCR 检测食品中 沙门氏菌 张体银,黄晓蓉,郑 晶,邵碧英,黄嫦娇,陈 彬,林 杰,汤敏英 (福建出入境检验检疫局,福建 福州 350001) 摘 要:利用免疫吸附富集结合经典 PCR 技术建立免疫捕捉通用引物 PCR(IC-UPPCR)检测食品中沙门氏菌。采用 细菌 16S rRNA 基因保守区设计特异性引物,建立通用引物 PCR 技术是可行的,该 IC-UPPCR 检测沙门氏菌的灵敏 2 度最低,能检测到 2 ×10 CFU/ml ,检测沙门氏菌属和非沙门氏菌属标准株的特异性为 100%,无假阳性和假阴性 出现。结果表明该方法具有简单、快速、特异性好和敏感性高等特点,并可满足大批样品沙门氏菌筛选检测的要 求,适用于食品卫生监管、商品检验检疫以及临床诊断等领域。 关键词:免疫捕捉;通用引物;P C R ;沙门氏菌;食品 Immunocapture-Universal Primer PCR Detection of Salmonella sp. in Foods ZHANG Ti-yin ,HUANG Xiao-rong ,ZHENG Jing ,SHAO Bi-ying,HUANG Chang-jiao , CHEN Bin ,LIN Jie ,TANG Min-ying (Fujian Entry-Excit Inspection and Quarantine Bureau, Fuzhou 350001, China) Abstract :An immunocapture-universal primer polymerase chain reaction (PCR) (IC-UPPCR) method was developed by combining the floride ion-selective electrode method with classical PCR assay for detection of Salmonella sp. in food and its sensitivity and specificity were investigated. It was feasible to establish a universal primer PCR technology was by designing specific primers from conserved regions of bacterial 16S ribosomal RNA genes. The sensitivity of the IC-UPPCR to detect Salmonella sp. was 2 × 102 CFU/ml and the specificity was 100% without any false negative or positive cases. The results demonstrate that the IC-UPPCR is a simple, rapid, highly sensitive and specific method for the detection of Salmonella sp. in the foods and suitable for detection of Salmonella sp. in a large number of food samples.
