南开大学硕 卜研究生毕业 (学位)论文 摘 要 基质金属蛋白酶 (MMPs)是一组降解连接组织的蛋白酶，它们的异常表 达可增加细胞外恭质和血管基膜的降解，促使肿瘤细胞的侵润和转移。为了弄 清基质余属蛋白酶 (MMPs)及其组织抑制因子 (TIMPs)在前列腺肿瘤的发生 及侵润过程中的作用。我们用RT-PCR的方法研究了MMP2,MMP7,TIMPI, TIMP2,MT1-MMP和MT2-MMP在前列腺组织和体外培养的各种类型的前列 腺细胞中的表达:用酶谱电泳的方法检测了MMP2,MMP9在人外周血中的含 量。 结果表明，MMP7在前列腺癌组织中的表达明显高于前列腺增生及正常 组织;MMP7ITIMPI比值的差异与前列腺肿瘤的发生和侵润高度相关:MMP9 在前列腺癌患者外周血中的含量高于增生患者，而其在增生患者外周血的含量 又高于正常患者。 结论:MMP7和MMP9是前列腺癌发生和转移的重要指标。 关键词:前列腺癌 基质金属蛋白酶 组织抑制因子 R〕-PCR 南开大学硕士研究生毕业 (学位)论文 Abstract Matrixmetalloproteinases(MMPs)areagroupofproteinasesresponsiblefor thebreakdownofconnectivetissues,whoseabnormalexpressionmayincreasethe degradationofextracellularmatrixandbloodvesselbasilemmaandenhancethe invasionandtransferenceoftumorcells.Inordertodeterminethefunctionsof MMPsandtheirtissueinhibitors(TIMPs)inthedevelopmentandinvasionof prostatetumors,weusedaRT-PCRtechniquetoinvestigatetheexpressionof MMP2,MMP7,TIMP1、TIMP2,MTI-MMPandMT2-MMPinprostatetissue andvariousprostaticcelltypesculturedinvitro;Wealsoexaminedthecontentsof MMP2andMMP9 inhumanperipheralbloodbymeansofenzymaticspectrum electrophoresis TheresultsindicatethattheexpressionofMMP7inprostatecancertissueis obviouslyhigherthaninthenormalorhyperplasticprostatetissue;theratioof MMP7/TIMP1ishighlycorrelatedwiththedevelopmentandinvasionofprostate tumors;thecontentofMMP9intheperipheralbloodofpatientsdiagnosedwith prostatecancerishigherthanintheperipheralbloodofpatientswithprostate hyperplastic,anditscontentintheperipheralbloodofpatientswithBPHishigher thanintheperipheralbloodofthenormalperson.
