Research on Regulating the Proliferation and Apoptosis of Vascular Adventitial Fibroblasts in Hypertension by Antihypertensive Formula of Clearing Liver Fire and Supplementing Qi Specialty: Formulae of Traditional Chinese Medicine Author: Ren Chongjing Tutor: Professor Han Tao Abstract Objective: To investigate the protective effect of serum containing antihypertensive formula of clearing liver fire and supplementing qi on the proliferation model of VAF induced by Ang Ⅱ, the paper aims to probe into the molecular mechanism on vascular remodeling in hypertension, so as to find effective therapeutic methods for the treatment of hypertension. Methods: The VAF model of the proliferation induced by Ang Ⅱ is simulated. The examined index and methods are as follows: the influence on VAF proliferation through MTT method, the growth cycle and apoptosis of VAF observed by flow cytometric analysis, the c-myc gene expression in the cells by immunocytochemical method. Results: The serum containing antihypertensive formula of clearing liver fire and supplementing qi can decrease VAF proliferation and block the transform from G0/G1 period to synthesis of DNA period. The results show the c-myc expression reduced significantly. And it can increase apoptotic rate significantly. Conclusions: The serum containing antihypertensive formula of clearing liver fire and supplementing qi can improve the vascular remodeling. The mechanism maybe have a link to the inhibiting of the proliferation and the promoting the apoptosis of VAF.
