[Abstract] Objective:To explore the differential regulation of mouse fetal liver stem cells to mature hepatocytes after induced by recombinant adenovirus-mediated mouse bone morphogenetic proteins 9 (Adv-mBMP9). Methods:The mouse fetal liver stem cells infected by the constructed recombinant adenovirus expressing BMP9、Hepatocyte Growth Factor (HGF) 、and green fluorescent protein (GFP) . To investigate the regulation of stem cells differentiation, alpha fetoprotein (AFP), albumin (ALB) and cell keratin 18 (CK18) were tested by half quantitative PCR, Real-Time PCR, cellular immune fluorescence and fluorescent element enzyme gene report. Results: Compared to the GFP control group, the expression of AFP induced with BMP9 and HGF 7days later was decreased tested by half quantitative PCR and Real-Time PCR. On the contrary, the expression of ALB was increased. The specific marker of mature hepatocyte, ALB and CK18 have strong expression after 7 days induction which were tested by cellular immune fluorescence assay, and the negative result were observed with the GFP control group. The expression of ALB after 1d, 4d and 7d induction with BMP9 and HGF in stem cells was significantly increased compared to GFP control group tested by fluorescent element enzyme gene report.
