Effect of Different Nutrient Levels on the IMF Related Gene mRNA Expression in Mongolia Sheep Abstract This experiment was conducted to research the effects of nutrition restriction and compensation on the expression of IMF related gene and meat quality in Mongolian sheep. Eighty WuZhuMuQin sheep(average weight 14.72 kg) were assigned randomly to 1 of 4 groups: control group(CG); restricted groupⅠ(RGⅠ); restricted groupⅡ(RGⅡ) and restricted group Ⅲ(RGⅢ) respectively. The results were showed as follow: 1. A real-time PCR method which was suitable for detecting the mRNA expression of IMF related genes in Mongolian sheep was established. The method has good specificity, high sensitivity, quantitative accuracy, can be applied to subsequent trials quantitative detection of gene expression analysis. 2. The nutrition restriction promoted the expression of H-FABP、FAS and HSL gene. Low energy levels promoted the expression of LPL and PPAR gene, and low protein levels promoted the PPAR gene expression, but inhibited LPL gene expression. Both energy and protein restrition resulted in the significant decrease in the expression of LPL and PPAR genes. During the nutritional compensation period, the expression of H-FABP、FAS and PPAR genes of RG3 were significantly higher than those of CG, while the expression of LPL and HSL genes of each restriction group is lower than those of CG, indicated that the nutrition compensation had different effects on the IMF related genes. 3. At the end of the restriction period, the crude fat content, intramuscular fat content, back fat thickness and the shear force values of RG3 were significantly lower than those of CG, except the pH45. Protein restriction resulted in the significant increase in intramuscular fat content. After nutrition compensation, there were no significant differences between crude fat content, back fat thickness, pH24 and water loss rate.
