探討臺灣藜多醣調節巨噬細胞之免疫活性mmunomodulatory activity of polysaccharides isolated from Chenopodium formosanum Koidz in macrophage model 研究生:施品岑 Shih, Pin-Tsen 指導教授:蔡碧仁 Tsai, Pi - Jen 【摘要】 巨噬細胞為高吞噬性功能細胞廣泛分佈於全身各處,並在先天性免疫反應和發炎反應中扮演重要角色,本研究使用臺灣藜(Chenopodium formosanum Koidz)種子萃取其多醣,並以小鼠巨噬細胞之免疫平台進行免疫活性及其探討。臺灣藜臺灣藜多醣NF-κB p65從細胞質轉位至核內,進而調節巨噬細胞之活性。進一步探討抗原呈分子與共刺激因子,發現隨著臺灣藜多醣濃度提高皆能增加細胞膜表面抗原呈獻分子第二類主要組織相容性複合物(major histocompatibility complex class II, MHCII)與共刺激因子蛋白(cluster of differentiation, CD80/86)之表現量。臺灣藜多醣。臺灣藜、多醣、巨噬細胞、免疫調節、第二類主要組織相容性複合物 【Abstract】 Macrophages are phagocytic cells widely distributed throughout the body, and play an essential role in innate immunity and inflammatory responses. Many polysaccharides isolated from plants are considered to be biological response modifiers and have been shown to enhance various immune responses in vivo and in vitro. In the present study, Djulis polysaccharide was isolated from Chenopodium formosanum seeds. It’s immune activity in mouse microphage model was investigated. We found that Djulis polysaccharide induced morphological changes of macrophages. Various cellular activities, such as phagocytosis, ROS generation, increased expression of inducible nitric oxide synthase iNOS , and NO production were observed in Djulis polysaccharide treated macrophages. Additionally , modulation of macrophage function by Djulis polysaccharides was mediated, at least in part, through activation of nuclear factor κB. Djulis polysaccharide exhibits specific funCtions in activating the major histocompatibility complex class II MHCII and co-stimulatory molecules, cluster of differentiation CD 80/CD86. These data suggest that Djulis polysaccharide may have regulatory functions on macrophage-mediated immune responses and support the concept of using Djulis polysaccharides as an immunotherapeutic adjuvant.
