结直肠癌组织中Skp2和P27kip1蛋白的表达及其临床意义 　　【摘要】目的探讨Skp2、P27kip12蛋白在结直肠癌中的表达及相关性，揭示Skp2和P27kip12在结直肠癌的发生和发展中的作用。方法应用免疫组织化学SP法检测Skp2、P27kip12、p53、bcl-2蛋白在100例结直肠癌中的表达情况。结果Skp2在结直肠癌组织中表达阳性率为64.0%（64/100），在正常组织中表达的阳性率为15.0%（6/40）；P27kip12在结直肠癌组织中表达阳性率为22.0%（22/100），在正常组织中表达的阳性率为90.0%（36/40）；p53在结直肠癌组织中表达阳性率为80.0%（40/50），在正常组织中表达的阳性率为0.0%（0/20）；结直肠癌组织中bcl-2蛋白阳性表达率为74.0%（74/100），显著高于结肠正常组织10.0%（4/40）（P0.05）。结论Skp2、P27kip12基因的异常表达是结直肠癌组织的发生、发展的原因之一。癌基因p53 及凋亡抑制基因Bcl-2的表达上调是其作用机制之一。Skp2和P27kip12基因的异常表达参与了结直肠癌的病理分化程度、临床Dukes分期、淋巴结转移。 　　【关键词】结直肠癌；Skp2；P27kip12；p53；bcl-2 　　Skp2 in Colorectal Carcinomas and Expression of P27kip12 Protein and Its Clinical SignificanceSUN Xiao.（Fengtian Hospital of Shenyang Medical College， Shenyang 110024，China）【Abstract】ObjectiveTo investigate the expression and correlation of Skp2 and P27kip12 in colon carcinoma and detect the relationship with the development of the diseases.MethodsTo detect the protein expression of Skp2 and P27kip12 by immunohistochemistry staining （SP method） in 100 cases of colon cancer and 40 normal colonic mucosas.Resultsthe positive expression of Skp2 and P27kip12 mainly located within the cellular nucleus. The protein expression of skp2 was 64.0%， significantly higher than normal colonic mucosa. 15.0% （P0.05）. The protein expression of P27kip12 was 22.0% in the colonic cancer， significantly lower than normal colonic mucosa. 90.0% （P0.05）. The positive expression of p53 is no expression in the normal colon tissue（0.0%， 0/40）， but there was significant increased expression in the colon cancer（80.0%， 80/100， P0.01）. There was a significant increase of positive expression of bcl-2 in the colon cancer 74.0%（74/100）compared with the normal tissue10.0%（4/40）（P0.05）. ConclusionsThe abnormal expression of both Skp2 and P27kip12 is one of the reasons that cause the development of colon cancer.
