2572 anti-fungal interleukin-6 receptor activity constituents
Author: Zou Ying-shu Zhang Yang Jiang Rong Li Jian-Bo Wu
【Abstract】 Objective 2572 metabolites from fungi isolated interleukin-6 receptor (IL-6R) antagonist. Methods using multiple chromatographic techniques for separation and purification of the active site and, based on MS, NMR and other spectroscopic methods the structure of the compounds were identified. The results from the fungal metabolites of the active site of 2572 to get two compounds, 2572A and 2572B. Conclusion 2572A is known as the 6-epi-5’-hydroxymycosporulone, with IL-6R antagonist activity, 2572B for riboflavin, active weak.

ABSTRACT Objective To isolate IL-6R antagonists from the cultured broth of the fungus 2572. Methods The compounds were separated by multi-chromatography and their chemical structures were elucidated by spectral analysis. Results Two known compounds, 2572A and 2572B were purified from the fermented extracts . Conclusions 2572A and 2572B were identified to be 6-epi-5’-hydroxymycosporulone and riboflavin, respectively. And 2572A shows IL-6Ra activity whereas 2572A has weak activity.
KEY WORDS Human interleukin 6 receptor antagonist; 6-epi-5’-hydroxymycosporulone; Riboflavin; Structure identification
Interleukin-6 (IL-6) is a feature a wide range of cytokines, and its biological function is very complex and in a variety of diseases such as rheumatoid arthritis, Castleman’s disease, multiple myeloma embolism play an important role [1]. IL-6 through the formation of IL-6/IL-6R/gp130 hexamer complex before they are allowed signal transduction, to play its biological activity, while interleukin-6 receptor (IL-6R) is mediated by the hexamer complex formed an important intermediary molecule [2]. In this study, selected to recombinant interleukin-soluble receptor (
