Results:BALB/cmicewereintraperitoneally(i.p)injectedwithcoxsackievirusB3(CVB3)forestablishingVMC models.Controlmiceweretreatedwithphosphate-bufferedsalinei.p.Onday0,7,14,21,28,35,42afterinjection, myocardialhistopathologicalchangeswereevaluatedbyhematoxylin-eosinstaining.SplenicTh17andTh9cells subsetswereanalyzedbyflowcytometry.AndcardiacIL-17,IL-9mRNAweremeasuredbysemi-quantitative reversetranscription-PCRandnestedPCR,respectively.ResultsshowedthelevelsofTh17cellsandIL-17mRNA obviouslyincreasedinVMCmiceon7dayafterinfection,peakedonday28,andhighlypersistedtoatleastday 42(p0.05).WhilethefrequenciesofTh9cellsandIL-9mRNAshowednosignificantdifferencebetweenVMC andcontrolgroupthroughoutthecourseoftheexperiment(p0.05). Conclusions:ItwasdifferentiatedTh17butnotTh9cellssignificantlyelevatedinthedevelopmentofCVB3- inducedVMC.ThemicroenvironmentofVMCseemedtocontributetothedifferentiationandproliferationofTh17 ratherthanTh9cells.OurpreliminarydataimpliedTh9cellscouldnotprotectagainstVMCnorpromotethe disease. Background BesidesTh1,Th2andTregcells,ourpreviousreportand Viral myocarditis(VMC) isanimportant cause ofheart otherresearchesdemonstratedTh17cellswhichprefer- failure inyoungadults andoften progresses tochronic entially secret cytokine IL-17 might play an important myocarditis, dilated cardiomyopathy(DCM) requiring role in mediat
