Toevaluatetheperformanceofthemagneticnanoparticlesasgenetransfervectorforbreedingtransgenicanimals,weinvestigated a new approach to deliver green ?uorescent protein (GFP) gene to porcine kidney 15 (PK-15) and porcine embryonic ?broblast (PEF) cells using PEI-modi?ed magnetic nanoparticles as gene vector. The morphology of the nanoparticles and nanoparticle/DNAcomplexeswascharacterizedusingscanningelectronmicroscopy.Itwasfoundthatthesurfaceoftheparticles becomes coarse and rough with increased average diameter, which implied the e?ective conjugating between nanoparticles withDNA.Thezetapotentialofnanoparticle/DNAcomplexesdropsdownfrom+29.4mVto+23.1mVcomparingwithpure nanoparticles.AgarosegelelectrophoresisexperimentsshowthatDNAplasmidscanbeprotectede?ectivelyagainstdegradationof exonucleaseandendonuclease.Thee?ciencyofgenedeliverywasa?ectedbythemassratioofnanoparticle/DNAandtheamount ofnanoparticle/DNAcomplexes.Wecon?rmthatthemostoptimalmassratioofnanoparticle/DNAis1:1b
