ThegeneticsandbiochemistryoftheN-linkedglycosylationsystemofArchaeahavebeeninvestigatedoverthepast5yearsusing ?agellinsandSlayersasreporterproteinsinthemodelorganisms,Methanococcusvoltae,Methanococcusmaripaludis,andHaloferax volcanii.StructuresofarchaealN-linkedglycanshaveindicatedavarietyoflinkingsugarsaswellasuniquesugarcomponents.In M.voltae,M.maripaludis,andH.volcanii,anumberofarchaealglycosylationgenes(agl)havebeenidenti?edbydeletionand complementationstudies.Theseincludemanyoftheglycosyltransferasesandtheoligosaccharyltransferaseneededtoassemble theglycansaswellassomeofthegenesencodingenzymesrequiredforthebiosynthesisofthesugarsthemselves.TheN-linked glycosylationsystemisnotessentialforanyofM.voltae,M.maripaludis,orH.volcanii,asdemonstratedbythesuccessfulisolation of mutants carrying deletions in the oligosaccharyltransferase gene aglB (a homologue of the eukaryotic Stt3 subunit of the oligosaccharyltransferasecomplex).However,mutationsthata?ecttheglycanstructurehaveseriouse?ectsonboth?agellation andSlayerfunction. 1.Introduction attachedbybothOandNlinkageoccur[7].Extremelylittle isknownoftheO-linkedprocessinArchaea. N-linked glycosylation is one of the most common post- translational modi?cations found on proteins in eukary- otic cells [1] and has now
