Developingvehiclesforthedeliveryoftherapeuticmolecules,likesiRNA,isanareaofactiveresearch.Nanoparticlescomposed ofbovineserumalbumin,stabilizedviatheadsorptionofpoly-L-lysine(PLL),havebeenshowntobepotentiallyinertdrug- deliveryvehicles.Withtheprimarygoalofreducingnonspeci?cproteinadsorption,thee?ectofusingcomb-typestructuresof poly(ethyleneglycol)(1kDa,PEG)unitsconjugatedtoPLL(4.2and24kDa)onBSA-NPproperties,apparentsiRNAreleaserate, cellviability,andcelluptakewereevaluated.PEGylatedPLLcoatingsresultedinNPswithζ-potentialsclosetoneutral.Incubation withplatelet-poorplasmashowedthecompositionoftheadsorbedproteomewassimilarforallsystems.siRNAwase?ectively encapsulatedandreleasedinasustainedmannerfromallNPs.With4.2kDaPLL,cellularuptakewasnota?ectedbythepresence ofPEG,butPEGcoatinginhibiteduptakewith24kDaPLLNPs.Moreover,24kDaPLLsystemswerecytotoxicandthiscytotoxicity wasdiminishedup
