AsymmetricDivergenceinStructureandFunctionof HCNChannelDuplicatesinCionaintestinalis HeatherA.Jackson1,AndrewHegle1,HamedNazzari1,TimothyJegla2,EricA.Accili1* 1Department of Cellular and Physiological Sciences, University of British Columbia, Vancouver, British Columbia, Canada, 2Department of Biology, Eberly College of Science,PennsylvaniaStateUniversity,UniversityPark,Pennsylvania,UnitedStatesofAmerica Abstract Hyperpolarization-activated Cyclic Nucleotide (HCN) channels are voltage-gated cation channels and are critical for regulationofmembranepotentialinelectricallyactivecells.Tounderstandtheevolutionofthesechannelsatthemolecular level, we cloned and examined two of three HCN homologs of the urochordate Ciona intestinalis (ciHCNa and ciHCNb). ciHCNaislikemammalianHCNsinthatitpossessessimilarelectricalfunctionandundergoesN-glycosylationofasequon nearthepore.ciHCNblacksthepore-associatedN-glycosylationsequonandispredictablynotN-glycosylated,anditalso has anunusual gating phenotype inwhich thechannel’svoltage-sensitive gateappearstocloseincompletely. Together withpreviousfindings,thedatasupportanevolutionarytrajectoryinwhichanHCNancestorunderwentlineage-specific duplicationinCiona,toyieldoneHCNwithmostfeaturesthatareconservedwiththemammalianHCNsandanotherHCN thathasbeenuniquelyaltered.
