Dysregulationofautophagyimpactsnumeroushumandiseasesincludingcancerandchronicobstructivelungdisease.This studyinvestigatestheroleofautophagyinidiopathicpulmonaryfibrosis. Methods:HumanlungtissuesfrompatientswithIPFwereanalyzedforautophagymarkersandmodulatingproteinsusing westernblotting,confocalmicroscopyandtransmissionelectronmicroscopy.TostudytheeffectsofTGF-b1onautophagy, humanlungfibroblastsweremonitoredbyfluorescencemicroscopyandwesternblotting.Invivoexperimentsweredone usingthebleomycin-inducedfibrosismousemodel. Results: Lung tissues from IPF patients demonstrate evidence of decreased autophagic activity as assessed by LC3, p62 proteinexpressionandimmunofluorescence,andnumbersofautophagosomes.TGF-b1inhibitsautophagyinfibroblastsin vitroatleastinpartviaactivationofmTORC1;expressionofTIGARisalsoincreasedinresponsetoTGF-b1.Inthebleomycin model of pulmonary fibrosis, rapamycin treatment is antifibrotic, and rapamycin also decreases expression of a′-smooth muscleactinandfibronectinbyfibroblastsinvitro.Inhibitionofkeyregulatorsofautophagy,LC3andbeclin-1,leadstothe oppositeeffectonfibroblastexpressionofa′-smooth muscleactinandfibronectin. Conclusion:Autophagyisnotinducedinpulmonaryfibrosisdespiteactivationofpathwaysknowntopromoteautophagy. ImpairmentofautophagybyT
