PrincipalFindings:Toevaluatetherespectivecontributionoftheseviralnucleicacidsinthereplicationoffoamyviruses, theirfatewasstudiedbyreal-timePCRandRT-PCRearlyafterinfection,inthepresenceorintheabsenceofAZT.Wefound thatanearlyreversetranscriptionstep,whichoccursduringthefirsthourspost-entry,isabsolutelyrequiredforproductive infection.Remarkably,sensitivitytoAZTcanbecounteractedbyincreasingthemultiplicityofinfection(moi).Wealsoshow that2-LTRcircularviralDNA,whichappearsassoonasfourhourspost-infection,istranscriptionallycompetent. Conclusion:Takentogether,ourdatademonstratethatanearlyreversetranscriptionprocess,whichtakesplacesoonafter viralentry,isindispensableforinfectivityofFVsatlowmoi,whentheamountofDNA-containingparticlesisnotsufficientto leadtoaproductiveinfection.ThisstudydemonstratesakeyroleofthepackagedviralRNAinthefoamyvirusinfection, suggestingthatthereplicationofthisviruscanbeachievedbyinvolvingeitherviralDNAorRNAgenome,dependingon theconditionofinfection.
