Bylabelingquantumdots(QDs)totheepidermalgrowthfactor(EGF),wecontinuouslytrackedtheendocytosisandpost- endocytic trafficking of EGF receptors (EGFRs) in A549 cells for a long time interval. A single-cell analysis method was introducedtoquantitativelystudythedynamicsofendocytictrafficking.Comparedwiththecontrolcells,thevelocityof directedmotionwasreducedby30%duetothesuppressionofhighspeedmovementsofEGF-QDsalongthemicrotubules inPTX-treatedcells.TheendocytictraffickinginPTX-treatedcellswasmainlyviasuper-diffusivemodeofmotion,whereasin controlcells,itwasmostlyviasub-diffusivemodeofmotion.Moreover,PTXshortenedendosomaltraffickingandprevented EGF-QDsfrommovingtotheperinuclearareaviatherapiddeliveryofEGF-QDsintotheperipherallysosomes.Thepresent studymayshedlightonthemechanismoftheeffectofPTXonthetreatmentoflungcancer.
