EfficiencyofOrganelleCapturebyMicrotubulesas aFunctionofCentrosomeNucleationCapacity:General TheoryandtheSpecialCaseofPolyspermia IvanV.Maly* DepartmentofComputationalandSystemsBiology,UniversityofPittsburghSchoolofMedicine,Pittsburgh,Pennsylvania,UnitedStatesofAmerica Abstract Transportoforganellesalongmicrotubulesisessentialforthecellmetabolismandmorphogenesis.Thepresentedanalysis derivestheprobabilitythatanorganelleofagivensizecomesincontactwiththemicrotubuleaster.Thequestionisaskedhow thismeasureoffunctionalityofthemicrotubuleasteriscontrolledbythecentrosome.Aquantitativemodelisdevelopedto addressthisquestion.Itisshownthatforthegivensetofcellularparameters,suchassizeandtotaltubulincontent,acentrosome nucleationcapacityexiststhatmaximizestheprobabilityoftheorganellecapture.Thedevelopedgeneralmodelisthenapplied tothecaptureofthefemalepronucleusbymicrotubulesassembledonthespermcentrosome,followingphysiologically polyspermicfertilization.Thisapplicationhighlightsanunintuitivereflectionofnonlinearityofthenucleatedpolymerizationof thecellularpooloftubulin.Thepredictionthatthespermcentrosomeshouldloweritsnucleationcapacityinthefaceofthe competitionfromtheotherspermisastarkillustrationofthenewoptimalityprinciple.Overall,themodelcallsattentiontothe capabilitiesofthecentrosomalpathwayofregulationofthetransport-relatedfunctionalityofthemicrotubulecytoskeleton.It establishesaquantitativeandconceptualframeworkthatcanguideexperimentdesignandinterpretation.
