Development of recombinant BCG (rBCG) over-expressingpromisingimmunodominantantigensofM.tuberculosisrepresentsoneofthepotentialapproachesforthe developmentofvaccinesagainstTB. Methods/PrincipalFindings:ArecombinantstrainofBCG-rBCG85C,overexpressingtheantigen85C,asecretoryimmuno- dominantproteinofM.tuberculosis,wasevaluatedforitsprotectiveefficacyinguineapigsagainstM.tuberculosischallenge byaerosolroute.ImmunizationwithrBCG85Cresultedinasubstantialreductioninthelung(1.87log10,p,0.01)andspleen (2.36log10,p,0.001)bacillaryloadwithacommensuratereductioninpathologicaldamage,whencomparedtotheanimals immunizedwiththeparentBCGstrainat10weekspost-infection.rBCG85Ccontinuedtoprovidesuperiorprotectionover BCGevenwhenpost-challengeperiodwasprolongedto16weeks.Thecytokineprofileofpulmonarygranulomasrevealed thatthesuperiorprotectionimpartedbyrBCG85Cwasassociatedwitht
